人缺氧誘導因子-1α(HIF-1α)elisa試劑盒等ELISA試劑盒操作技術相關的注意事項:
⑴ 嚴格按照人缺氧誘導因子-1α(HIF-1α)elisa試劑盒說明書進行操作。操作前將試劑在室溫下平衡30~60min。
⑵ 加樣后及時放人孵箱。標本較多時,要分批操作。按說明步驟嚴格控制操作時間,防止孵育時間人為延長,導致非特異性結合緊附于反應孔周圍,難以清洗徹底。
(3)封板溫育時,各孔一定要封嚴,防止陽性標本的液體蒸發,產生周邊現象從而導致“花板”的出現。
(4)用洗板機洗板時,保證洗液注滿各孔,洗板針暢通,洗完板后最好在吸水紙(選擇干凈、無或少塵的吸水材料)上輕輕拍干:還要防止針口有纖維蛋白或異物,這些異物在洗板的過程中易產生拖帶現象而導致“花板”的出現。
(5)合理安排檢測量,以免反應板過多造成洗板等待時間長。
(6)加酶試劑后用吸水紙在酶標板表面輕拭吸干。
(7)顯色劑盡量在臨用前配制,堅持不用過期顯色劑,肉眼可見淺藍色的TMB顯色劑不用。
(8)加樣時保持顯色劑不外流;A、B液應避免接觸金屬器械。加終止液時應避免產生氣泡。
(9)應保證C清潔,整個操作過程中保證酶標板不接觸次氯酸。以上的措施可以使“花板”降至最低限度。從而提高檢測的特異性。并得到更準確、可靠的實驗結果。
